Expression and correlation of interlukin-1β, tumor necrosis factor-α and matrix metalloproteinase-13 in the synovial fluid of osteoarthritis patients

Abstract
Objective To investigate the expression of interlukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) and matrix metalloproteinase-13 (MMP-13) in the synovial fluid of osteoarthritis (OA) patients and their correlations with OA.Methods The expression of IL-1β, TNF-α and MMP-13 in the synovial fluid of 72 OA patients and 32 normal persons were assessed by RT-PCR and ELSIA.The severity of OA was accessed by the Hospital for Special Surgery (HSS) score.The pairwise correlations between IL-1β, TNF-α or MMP-13 and HSS score were analyzed by Pearson correlation analysis.Results The mRNA and protein levels of IL-1β, TNF-α and MMP-13 of OA patients were all significantly higher than those of normal persons (P<0.01).There were significant positive correlations of IL-1β, TNF-α and MMP-13 with the severity of OA.The levels of IL-1β and TNF-α had significantly positive correlation with MMP-13 in OA patients.Conclusion IL-1β, TNF-α and MMP-13 are closely related to the development and progression of OA, as well as correlated to the severity of OA.The high expression of MMP-13 in the synovial fluid may be activated by IL-1β and TNF-α.It is important to measure the levels of IL-1β, TNF-α and MMP-13 in the synovial fluid for the diagnosis, evaluation and treatment of osteoarthritis.关键词
