目的 构建靶向TPX2基因的RNA干扰慢病毒载体,获得稳定感染的人宫颈癌HeLa细胞株,为宫颈癌细胞TPX2基因的相关研究奠定基础.方法 以TPX2基因为靶点,设计并合成四组对应于目标shRNA的双链DNA发卡结构,分别与慢病毒载体Pglv2-U6-Puro连接经转化感受态细胞,阳性克隆经测序、病毒包装、滴度测定、感染细胞、嘌呤霉素筛选得到TPX2基因沉默稳转细胞株.实时荧光定量PCR和Western blot检测TPX2 mRNA和蛋白的沉默效…查看全部>>
