摘要

Abstract

In this study,eukaryotic expression plasmid pcDNA3.1(+)-CDNF(cerebral dopamine neurotrophic factor)was recombined to bone mesenchymal stem cell (BMSCs) by the Lipo 2000 for establishing CDNF-BMSCs.CDNF-BMSCs were transplanted into the striate body of the PD model rats for the therapy by brain stereotaxic technique.Rotating behavior was tested in different periods after treatment respectively.Immunohistochemical and WB technology were took to detect the expression of CDNF and tyrosine hydroxylase (TH) in the striate body for exploring the effect of the CDNFBMSCs on Parkinson's disease(PD).The result showed that the behavior of CDNF-BMSCs group was improved significantly after two weeks from transplantation treatment.CDNF protein expression was very significantly increased and TH protein expression was increased significantly.At the same time,the CDNF and TH positive cells in the striate body of CDNF-BMSCs group increased gradually over time.Therefore,this study provided an experimental evidence for the gene therapy of neurodegenerative diseases,such as Parkinson's disease.
