摘要

Abstract

The objectives of this study were to investigate the effect of curcumin on myocardial antioxidant,glucose metabolism and apoptosis in rats after exhaustive exercise and to explore the mechanism of myocardial protection in exhaustive rats.50 SD rats were randomly divided into 4 groups:control group (CK),exhaustive exercise control group (EE),low dose curcumin group (LC) and high dose curcumin group (HC).LC group and HC group treated with different doses of curcumin stimulation based on movement of rats in stress were detected for myocardial tissue malondialdehyde (MDA),ELISA and glutathione peroxidase (GSH-Px),catalase (CAT) and superoxide dismutase (SOD) activity.Western blot (WB) was used to detect the expression of key proteins in glucose metabolism,Bcl-2 and the expression of apoptosis related proteins.The results showed that compared with EE group,the activity of SOD,CAT and GSH-Px in myocardium of LC group and HC group were significantly higher (P<0.05),MDA content was significantly lower (P<0.05),the expression of Nrf2 was increased,and the expression of Glutl and HK2 was up-regulated.Compared with Control,the expression of Bcl-2 and Mcl-1 in the EE group was down regulated,and the expression of caspase3/7 and PARP increased.However,the Bcl-2 and Mcl-1 protein in LC group and HC group were significantly higher than those in EE group,and the expression of caspase3/7 and PARP decreased.Cureumin can enhance the antioxidant capacity of rat myocardial cells,promote glucose metabolism and inhibit the occurrence of apoptosis.
