Abstract
Objective:This study aims to understand the pathogenesis of endothelial-to-mesenchymal transition(EndMT)induced by arecoline and explore the change of vessel in OSF and possible pathogenesis of OSF by investigating the proliferation of human umbilical vein endothelial cells(HUVECs)and the expression of Ang-Ⅱ,TGF-β1 and α-SMA influenced by arecoline.Methods:The survival rate of HUVECs was observed that influenced by increasing concentrations of arecoline(0,2.5,5,10,20,40,80 μg/mL)and different time(24,48,96 h)detected by methyl-thiazol-tetrazolium(MTT)assay and flow cytometry.The expression of Ang-Ⅱ,TGF-β1 and α-SMA of HUVECs was observed that influenced by increasing concentration of arecoline and different time detected by Western Blot.Results:MTT shows that the survive rate of HUVECs decreases with the increaseing concentration of arecoline and the extension of time.Flow cytometry shows that HUVECs begin to apoptosis obviously when the concentration of arecoline was 20 μg/mL.And apoptosis rate increases with the increaseing concentration of arecoline or the extension of time.Ang-Ⅱ,TGF-β1 and α-SMA express increasing firstly and then decreasing,with the strongest expression lying on 96 h influenced by 10 μg/mL of arecoline.The differences are statistically significant(P<0.05).The variation trend was consistent with the concentration of arecoline and the extension of time.Conclusion:The HUVECs apoptosis induced by arecoline in vitro,and abnormal apoptosis of endothelial cells may be a possible pathogenesis of change of vessel in OSF.Arecoline can induce endothelial-to-mesenchymal transition of HUVECs.The endothelial-to-mesenchymal transition may be realted to the signal regulationg of Ang-Ⅱand TGF-β1.关键词
