Effect of different culture systems on in vitro development of mouse embryos
摘要
Abstract
Objective To investigate the effects of different culture media,different culture methods and culture gas on the in vitro development of mouse embryos.Methods The mouse embryos was divided into Cook group,Quinns group,Vitrolife group according to different brands of culture medium.The embryos were obtained by in vitro fertilization among three groups,and the fertilization rate,cleavage rate and blastocyst rate were compared.According to different culture methods,the experiment was divided into single embryo culture group,WOW group and multiple embryos group.The embryos were obtained by in vivo fertilization among these groups to compare cleavage rate and blastocyst rate.According to the culture phase,the experiment was divided into two gas group and three gas group.The embryos were obtained by in vivo fertilization in the two groups to compare the cleavage rate,blastocyst rate and Fragmentation rate.Results The fertilization rate was the highest in Quinns group(90.7%).There was no significant difference in cleavage rate among three groups (85.4%,84.0%,84.9%,P >0.05).The blastocyst rate was the highest in V itrolife group (89.6%),and there was no significant difference among three groups(P > 0.05).There was no significant difference in cleavage rate among three culture methods (P > 0.05).Blastocyst rates in multiple embryos group and WOW group were significantly higher than that of single embryo culture (89.1%,88.1% vs 77.5%,P < 0.05).There was significant difference in cleavage rate (74.7%,92.6%) and blastocyst rate (76.4%,90.9%) between two culture gas groups (P < 0.05).Conclusion The different culture medium,different culture methods and culture gas have different effects on mouse embryo development in vitro.The results suggest that Quinns medium as fertilization media,Vitrolife media as the following culture media,multiple embryos group culture method and three gas(6% CO2,5% O2and 89% N2) as culture environment can have higher blastocyst rate and lower fragmentation rate in vitro development of mouse embryos.关键词
