Screening and Validation of PAG1 Interacting Proteins

Abstract
Objective To screen and validate PAG1 (phosphoprotein associated with glycosphingolipid icrodomains 1) interacting proteins in the inherently radioresistant laryngeal cancer cells.Methods The binding proteins of PAG1 were enriched using immunoprecipitation(IP) assay with anti-PAG1 specific antibody.The immunoprecipitates were separated by SDSPAGE.Then,the binding proteins were cut from the gel and analyzed by mass spectrometry (MS).The function of each protein was obtained from the NCBI.In addition,the immunofluorescence double labeling assays were used to verify the interacting protein.Results Totally,9 potential PAG1 partners,including integrin β1,were obtained by the method of IP coupled with MS.The colocalization of PAG1 and integrin β1 was confirmed by immunofluorescence double labeling assay.Conclusion PAG1 may interact with integrin β1 in the inherently radioresistant laryngeal cancer cells,which lay the foundation for further study of PAG1 function.关键词
