细胞间黏附分子-1与小鼠新生血管的相关性研究
Relationship between intercellular cell adhesion molecule-1 and angiogenesis in mice

Abstract
Objective To study the correlation between the expression of intercellular cell adhesion molecule-1 (ICAM-1) and angiogenesis in atherosclerosis.Methods Eight week old ApoE-/-mice were fed with high fat diet up to 16,24,and 32 weeks respectively,to establish an experimental animal model for the formation of atherosclerotic lesions from the beginning to the fatty streaks,fibrous plaques and atherosclerotic plaques.Determination the total cholesterol (TC),low density lipoprotein (LDL),triglyceride (TG),high density lipoprotein (HDL) by automatic biochemical analyzer,ICAM-1 in atherosclerotic plaque was detected by immunohistochemistry.Immunohistochemistry labeling of neovascularization with CD31 antibody.Results TC and LDL increased gradually with the increase of age.The TC in 8,12,24,32 week mice were (1.48±0.13),(3.56±0.25),(8.95 ±0.33),(12.23 ±0.58) mmol · L-1.The LDL in 8,12,24,32 week mice were (0.65 ±0.06),(2.36±0.18),(4.19 ±0.31),(5.64 ±0.55) mmol· L-1,had significant difference compared with 8 week old mice (P < 0.05).The relative expression of ICAM-1 increased gradually with age.The ICAM-1 of 8,16,24,32 week old mice were 2.12 ± 0.56,7.17 ± 2.64,12.86 ± 3.72,16.78 ± 3.52.The number of new vessels of 8,16,24,32 week old mice were 2.12 ±0.56,7.17 ±2.64,12.86 ±3.72,16.78 ±3.52,had significant difference compared with 8 week old mice (P < 0.05).ICAM-1 was positively correlated with neovascularization (y =2.79x + 3.40,r2 =0.927 9).Concltsion In the process of atherosclerosis,the expression of ICAM-1 and angiogenesis gradually increased,and the increased expression of ICAM-1 may be the reason of the increase of new blood vessels.关键词
