Abstract

Three low content flavonoids, such as vitexin, hyperoside and isoquercitrin from Crataegi Folium were rapidly separated and purified by high-speed counter-current chromatography (HSCCC) combined with pre-HPLC.The total flavonoids from Crataegi Folium were firstly separated by HSCCC, with a solvent system of chloroform/methanol/water/ n-butanol (4∶3∶2∶1.5, V/V), flow-rate of 5.0 mL/min, rotation speed of 850 r/min, detection wavelength of 254 nm.A mixed component was separated from crude flavonoids, containing vitexin, hyperoside, and isoquercitrin.Then, the mixture was separated by pre-HPLC and vitexin, hyperoside, and isoquercitrin were obtained, with the purity over 98%, respectively, as determined by HPLC.Experimental results show that it is an efficient method for separation of low content flavonoids with high purities from Crataegi Folium by HSCCC combined with pre-HPLC.
