O型口蹄疫病毒不同宿主适应毒3A基因序列分析
Nucleotide and amino acid sequence analysis of 3A gene of foot and mouth disease virus type O in different hosts

Abstract
In order to analyze the nucleotide and amino acid sequences of the gene encoding 3A protein of Mya98 lineage foot and mouth disease virus (FMDV) type O adapted in different hosts and therefore viral evolution and variation of FMDV,FMDV strain O/X J/10-11 was inoculated in bovine,swine,rat and BHK-21 cells and the host-adapted FMDV strains were obtained.RNA in tissue fluid was extracted and used as the template for reverse transcription and amplification of 3A gene.Then these sequences were analyzed by Vector NTI 11.5 and DNAStar biological software.The results showed that in the four host-adapted virus strains from bovine,rat,swine and BHK-21 cells,3A gene was more stable,and variation occurred in aa 99 and aa 114,indicating that the two amino acids of FMDV 3A gene are related to host tropism to a certain extent.The results lay a foundation for further analysis of the variability of the genomes from the different host-adapted FMDV strains.关键词
