猪ZBED6基因有效突变位点的筛选与验证
Screening and Identification of Effective Mutation Site in Pig ZBED6 Gene

Abstract
The mutation of ZEBD6 can promote the transcription of IGF2 in skeletal muscle,which could enhance the cell proliferation and myotube formation.In this study,CRISPR-Cas9 was used to editing the pig ZEBD6 gene.Four mutation targets were screened by online software,pCas9/gRNA-ZBED6 plasmid was constructed by pCas9/gRNA vector.At the same time,pTYNE-ZBED6 plasmid was constructed to verify the mutation efficient.These plasmids were transfected to HEK293 cell line in the meantime.The fluorescence signal was observed and was used to judge the mutation efficient of the target sequence.The results showed that,the f target 1,which is located in 17-37 nucleotide acid,has the strongest fluorescence signal.It means that CRISPR-Cas9 has the highest cutting efficiency in this site.It is the effective mutation site in ZBED6 gene which can be edited by CRISPR-Cas9.关键词
