Abstract
Objective To investigate the expression of TPX2 in papillary thyroid carcinoma ( PTC) tissues and its correlation with clinical pathological parameters, as well as its effect on KAT cell proliferation. Methods 60 cases of PTC tissues and paired normal tissues were collected in our hospital during 2013-2016. The mRNA level of TPX2 was determined by qRT-PCR. KAT cells were transfected with special siRNA to inhibit TPX2 expression, and then cell proliferation, cell apoptosis and apoptosis-associated protein ( cleaved caspase 3, caspase 3 ) were tested by MTT, flow cytometry and Western blot. Results Compared to the paired normal tissues, TPX2 was highly expressed in tumor tissues at mRNA level ( 39/60) ( P<0.05) . Knockdown of TPX2 effectively reduced KAT cell growth and increased cell apoptosis. Meantime, apoptosis-related proteins cleaved caspase 3 and caspase 3 were up-regulated in TPX2-siRNA-treated group. Conclusion The expression of TPX2 is higher in PTC tissues. TPX2 is highly correlated with cell proliferation, which may be a potential target for treatment of thyroid carcinoma.关键词
