Abstract
Objective:To discuss the effect of acupuncture on NGF-TrkA and AKT for rats with chronic ocular hypertension.Methods:SD rats were randomly divided into controlled group,model group,treatment group,the model group and the treatment group were treated with compound carbomer induced rat model of chronic intraocular pressure.After 2weeks,the treatment group was given acupuncture at Jingming and Qiuhou acupuncture treatment;the controlled group and the model group were not treated.After treatment,5rats in each group were observed the morphological changes of the eye,retinal tissue HE staining,the transcription level of NGFmRNA,TrKAmRNA and p75mRNA was detected by RT-PCR;Western blotting method was used to detect the AKT (p-AKT) expression level of the retinal NGF signal pathway downstream molecule.Results:After modeling,compared with the controlled group,the model group and the treatment group had elevated intraocular pressure (P < 0.05);Compared with the model group,the treatment group at each time point had significant difference in intraocular pressure (P < 0.05).Compared with the controlled group,the transcription level of NGFmRNA in the model group and the treatment group increased (P < 0.05),and the treatment group was higher than the modelled group.Compared with the model group,the transcription of TrkA mRNA was significantly higher in the treatment group (P < 0.01).Compared with the controlled group,the p75mRNA level of the model group and the treatment.group was lower than that of the model group (P < 0.05),and the treatment group was lower than the model group.Compared with the controlled group,the expression of downstream signaling molecule AKT (p-AKT) in the treatment group increased,compared with the model group,the treatment group had an increasing trend.After acupuncture,the ganglion cell layer spacing decreases by increasing the number of nuclei,which could reduce the pathological damage of the retina,the stability of the number of ganglion cells,retinal ganglion cells in chronic high intraocular pressure in rabbits after the rat model of effective protection,and improve the survival rate.Conclusion:Acupuncture can protect RGCs by NGF TrkA signaling pathway,which provides a theoretical basis for acupuncture treatment of glaucoma.关键词
