摘要

Abstract

Objective:To investigate the effect of maternal gestational diabetes mellitus on glycometabolism of female offspring in SD rat.Methods:Sprague-Dawley rats (8-week-old) were used in this study.Maternal gestational diabetes mellitus was induced by a single injection of streptozotocin (STZ) on the first day of gestation in Sprague-Dawley rats.Control rats were injected with citrate buffer.Serum insulin and lipids were determined by enzyme linked immunosorbent assay (ELISA).Islet area were evaluated through haematoxylin and eosin staining.Glucose and insulin tolerance tests (GTT and ITT) and insulin releasing test (IRT) were performed.microRNAs expression were evaluated through qPCR in islet.Results:Compared with offspring control group,diabetes offspring group showed more slight impairment in GTT and ITT.In addition,they showed bigger islet throughout the study.The miRNA-130a expression was decreased in diabetes offspring group.Conclusion:Maternal gestational diabetes mellitus may be a high-risk factor of glycometabolism disturbance in female offspring.
