Abstract
Objective: To investigate the effects of piperine on SKOV-3 ovarian cancer cell line invasion and apoptosis and to explore the underlying mechanisms. Methods: SKOV-3 cells were treated with different concentrations of piperine (0, 5, 10 or 20 μmol/L) for different times (12, 24 or 36 h), cell viability was assessed by CCK-8 assay. The invasion and apoptosis of SKOV-3 cells were determined with Transwell and TUNEL staining, respectively. The protein expression of p-PI3K and p-Akt were examined by Western blot. Results: CCK-8 results showed that piperine decreased the viability of SKOV-3 cells in a dose-dependent and time-dependent manner. After treatment with 10 μmol/L of piperine for 36 h, cell invasion was significantly inhibited, whereas the apoptotic cell number was markedly increased. Moreover, the protein expression of p-PI3K and p-Akt were significantly decreased after piperine treatment. Conclusion: Piperine can inhibit SKOV-3 cell viability and invasion, and increase cell apoptosis, which may through inhibition of PI3K/Akt signaling pathway.关键词
