灯盏生脉胶囊对帕金森病大鼠黑质区多巴胺能神经元损伤影响
EFFECT OF DENGZHAN SHENGMAI CAPSULES ON DOPAMINERGIC NEURON INJURY IN SUBSTANTIA NIGRA IN RATS WITH PARKINSON'S DISEASE

Abstract
Objective To investigate the effect of Dengzhan Shengmai capsules on dopaminergic neuron injury in the substantia nigra in rats with Parkinson's disease (PD) induced by 6-hydroxydopamine (6OHDA),as well as its mechanism in DNA oxidative damage repair.Methods A total of 80 rats were randomly divided into sham-operated group,model group,low-dose Dengzhan Shengmai group (low-dose group),and high-dose Dengzhan Shengmai group (high-dose group).Injection of 6-OHDA into the striatum was performed to establish a rat model of PD.The rats in the low-and high-dose Dengzhan Shengmai groups were given low-or high-dose Dengzhan Shengmai capsules by gavage from 3 days before modeling to week 5 before sacrifice,and those in the sham-operation group and the model group were given normal saline at the same dose.The changes in rotational behaviors induced by apomorphine were observed at week 4 after modeling.After the rats were beheaded and brain tissue was collected at week 5,immunohistochemistry and Western blot were used to measure the number of cells with tyrosine hydroxylase (TH) or MTH1 in the substantia nigra and their protein expression levels.Results There was a significant difference in the rotation rate between the four groups (x2 =21.89,P<0.05).The number of cells with TH in substantia nigra pars compacta at the operation side was 78±13 in the sham-operation group,27±7 in the model group,39±8 in the low-dose group,and 54±11 in the high-dose group;there was a significant difference between the four groups (F =43.89,P <0.95).The high-dose group had a significantly higher content of TH protein in the substantia nigra at the operation side than the model group (F =9.50,P <0.05).The number of cells with MTH1 in the substantia nigra pars compacta at the operation side was 42±7 in the sham-operation group,22±6 in the model group,36±6 in the low-dose group,and 45±8 in the high-dose group;there was a significant difference between the four groups (F =22.65,P <0.05).The high-dose group had the highest MTH1 content in the substantia nigra at the operation side,while the sham-operation group had the lowest content,and there was a significant difference between these two groups (F =50.49,P <0.05).Conclusion Dengzhan Shengmai capsules can effectively alleviate dopaminergic neuron injury in the substantia nigra in rats with PD,and its mechanism is associated with the improvement in DNA damage repair.关键词
