Abstract
The gene sequence VP1-ICAM-2 was synthesized by using PAS(PCR-based accurate synthesis) method.The recombinant plasmid was obtained by introducing the synthetic gene VP1-ICAM-2 into the transfer vector pFast-Baclby using BamH Ⅰ and HindⅢ,and then the recombinant plasmidpFast-Bacl-VP1-ICAM-2 was transformed into DH10Bac competent cells for blue-white screening to obtain positive recombinant granules rBacmid-VPl-ICAM-2.The recombinant baculovirus rBac-VPl-ICAM-2 was obtained by transfection of positive recombinant granulocytes into Sf9 cells.The Western-blot showed that the recombinant protein was about 57 ku in size and could be recognized by A-type FMDV positive sera.The guinea pigs experiment showed that the fusion protein VP1-ICAM-2 induced FMDV specific humoral immune and cellular immune responses.This study result provides informatinon for the development of new FMD subunit vaccines.关键词
