Abstract

The purpose of this study was to compare three assays with respect to their ability to accurately detect BoHV-1 in biological samples,namely nano PCR,loop-mediated isothermalamplification (LAMP),and conventional PCR.All assays detected BoHV-1 nucleic acids and differentiated between BoHV-1-free and-contaminated bovine sera successfully.A rapid,specific,sensitive,and practical method of detecting BoHV-1 in bio-products is needed.In addition,the results were specific to BoHV-1:theamplification of samples containing the other pathogenic bovine viruses yielded negative results.The lowest detection threshold,2.23 × 102 copies/μL,was achieved by the nano-PCR assay.This assay was also the most effective in the detection of BoHV-1 in biological samples.The fastest detection was in 50 min,which was achieved by the LAMP assay.The field conditions suggest that nano-PCR is specific and sensitive to detect BoHV-1 in biological samples and may be used for quality control of biomaterials.
