Abstract

A diarrhea outbreak characterized by watery diarrhea and high mortality among neonates occurred in a pig farm in Jiangsu province in March 2016.To identify the pathogen that caused this diarrheal disease,six feces samples were collected and subjected to RT-PCR analysis.The results showed six samples were positive for PEDV,while all of them were negative for TGEV and PoRV.Then all the positive samples were processed and inoculated into the monolayers of Veto cells for virus isolation.Only one sample designated JSqd3/2016 could induce visible cytopathogenic effect(CPE)and the virus isolated from this sample was serially passaged to the 5th generation (JSqd3/2016-FS).After that,the Vero cells inoculated with JSqd3/2016 were tested by indirect immunofluorescence assay(IFA)and the result showed it could be specifically recognized by the monoclonal antibody(MAb)against the PEDV spike(S) protein.Also,the S gene and ORF3 gene of JSqd3/2016-F5 were amplified,cloned,sequenced and used for sequence analysis.The result showed JSqd3/2016 was more closely related to the currently circulating variant PEDV strains;its S gene had pronounced mutations compared with the classical strains and the deduced amino acid sequence similarity between S protein of JSqd3/2016-F5 and the circulating variant PEDV strains ranged from 93.0%to 93.7%.These results suggested JSqd3/2016 was a variant PEDV strain and it was the causative agent of this diarrheal outbreak.
