胸腺五肽在植物乳杆菌中的表达及其活性检测
Expression and characterization of thymopentin in Lactobacillus plantarum

Abstract
The thymopentin(TP5) gene was engineed based on Lactobacillus plantarum-optimized codon usage,and ligated into the Lactobacillus expression vector pSCPSP,which was expressed by exogenous gene and driven by superpromoter SCP.The recombinant plasmid pSCPTP5 was constructed and transformed into the porcine Lactobacillus plantarum strain LPI by electroporation.The positive recombinant transformant pSCPTP5 was obtained successfully through identification of genome by PCR method.Afterwards,TP5 was expressed in Lp-1 and detected by ELISA.The expression levels of TP5 in the cultural supernatant was determined by specific ELISA kit at 16 h,20 h,24 h and 28 h.After orally irrigated early-weanedKMmice with the recombinant L.plantarum every day for 14 consecutive days,thymus and spleen index,and TL-2 in the serum were measured to examine the influence of the recombinant bacteria in vivo.Finally,the TP5 was successfully expressed in Lp-1 and the optimal incubation time has been measured at 24 h.In addition,the recombinant TP5 could exerted significance influence on the thymus index and IL-2 of early-weaned BALB/c mice.In conclusion,TP5 could be expressed in L.plantarum and the recombinant TP5 possesses good biological activities.关键词
