Abstract
Objective:The purpose of this study was to cultivate and characterize the dog bone marrow derived mesenchymal stem cells(BMSCs)and define the biological features and osteogenic differentiation potential of BMSCs.This study could provide experimental evidence for the bone tissue engineering.Methods:Bone marrow was extracted from the dog femurs.BMSCs were then isolated by density gradient centrifugation and adherent cultured.Growth curve of cultured BMSCs was plotted.Furthermore,the biological features of BMSCs were investigated by flow cytometry.The osteogenic differentiation and adipogenic differentiation were detected in vitro.Results:Monoptychial heterogeneous cells were harvested.Flow cytometry results showed that 99.51% cells expressed CD29 and 99.97% cells expressed CD90 in the P3 BMSCs.BMSCs were able to differentiate into osteogenic and adipogenic cells.Conclusion:BMSCs isolated from the dog bone marrow can serve as ideal seed cells for bone tissue engineering.关键词
