Abstract

In order to investigate the stability and antifungal activities of acetone extracts on Juglans mandshurica,ultrasonic and UV spectrophotometry method were used to extract and determine the tannins content of acetone extracts on leaves and green peel of Juglans mandshurica.Taking the tannin degradation rate as the evaluation index,the effects of temperature and sunshine on the stability of acetone extracts were studied.Results showed that the optimal extraction conditions were the volume fraction of acetone 50% with water,ultrasonic temperature of 50 ℃,extraction time of 40 min,solid-liquid ratio of 1∶15,the tannins contents were 38.6 mg·g-1 and 35.4 mg·g-1 with leaves and green peel of Juglans mandshurica respectively.With the increase of temperature or strong sunshine,the degradation rate of tannin increased and the stability decreased.The stability of acetone extract of leaves was slightly higher than that of green peel.The cylinder-plate method was used to evaluate the antifungal effect for wood-rooting fungi of the acetone extracts on leaves and green peel of Juglans mandshurica.Results showed that acetone extracts of Juglans mandshurica had antifungal activities for wood-rotting fungi of both Gloeophyllum trabeum and Coriolus versicolor.MIC of the acetone extracts on leaves and green peel of Juglans mandshurica were both 1.50 g/L for Gloeophyllum trabeum,and both 2.00g/L for Coriolus versicolor.The antifungal activity for Gloeophyllum trabeum was better than that for Coriolus versicolor.The acetone extracts on leaves and green peel of Juglans mandshurica are suitable storage and use for neutral condition at room temperature and away from light.
