野菊花保肝胶囊抗乙肝病毒及抗酒精性肝损伤的作用
Effects of Yejuhuabaogan capsule on HBV and acute alcoholic liver inju-ry in mice
摘要
Abstract
Aim:To investigate the effects of Yejuhuabaogan capsule ( CBC) on HBV in vitro and acute alcoholic liver injury in vivo.Methods: HepG2.2.15 cells were divided into 9 groups,and were treated with 0.000,0.625,1.250, 2.500,5.000,10.000,20.000,40.000, and 80.000 mg/L CBC for 72 hours.CCK-8 assay was used to detect the cell pro-liferation.HepG2.2.15 cells were assigned into 6 groups and were treated with 0.000,0.125,0.250,0.500,1.000 mg/L CBC and 100 mg/L lamivudine,and ELISA was used to detect the secretion of HBsAg and HBeAg by HepG 2.2.15 cells on the 6th and 9th day after the treatment.Kunming mice were allocated into 6 groups:control group, model group, BP posi-tive control group(150 mg/kg), CBC groups with high, middle and low dosages(280, 140 and 70 mg/kg).The liver inju-ry model was prepared with the volume fraction of 53%ethanol by gavage .The levels of serum ALT and AST were deter-mined, and liver homogenate was prepared to detect SOD and MDA activity after 10 days.The liver was studied by patho-logical examination .Results:CBC reduced the contents of HBsAg and HBeAg secreted by HepG 2.2.15 cells on the 6th and 9th day(P<0.05).TC50 and TC0 of CBC were 5.014 and 0.231 mg/L,respectively.CBC reduced the serum levels of ALT and AST, and increased SOD activity, depressed MDA content(P<0.05).CBC could alleviate liver injury,especial-ly at the high and middle doses(P<0.05).Conclusion:CBC has anti-HBV effects as well as the protective effects on the alcoholic liver injury .关键词
