TCDD对大鼠睾丸间质细胞凋亡相关蛋白表达及ROS含量的影响
Effects of TCDD on expressions of apoptosis related proteins and ROS content in leydig cells of rats

Abstract
Aim:To explore the effects of TCDD on expressions of apoptosis related proteins and ROS content in leydig cells of rats.Methods:Leydig cells from rats were treated with different doses of TCDD (0,1,5,10,50 nmol/L) for 24 hours, and the proliferation inhibition rate of leydig cells was determined by MTT assay , ROS content in leydig cells was determined by DCFH-DA method, apoptosis was observed by confocal microscopy combined Annexin V -FITC/PI staining, and the expressions of Bax , Caspase-3 and Bcl-2 proteins were detected by Western blot .Results:With the increase of the dose of TCDD, the proliferation inhibition rate and ROS content showed a rising trend (P<0.05), the expressions of Bax and Caspase-3 increased whereas that of Bcl-2 protein decreased(P<0.05).Conclusion:TCDD could significantly induce apoptosis of rat leydig cells .关键词
