五参胶囊镇静催眠作用及其机制的实验研究
Sedative and Hypnotic Effects and Mechanisms of Wushen Capsules
摘要
Abstract
Objective To observe the sedative and hypnotic effects of Wushen Capsules ( WShC) and to explore the underlying mechanisms. Method Female ICR mice were randomly divided into 4 groups:control group,high-dose WShC group (200 mg/(kg·d-1)),medium-dose WShC group (100 mg/(kg·d-1)),and low-dose WShC group (50 mg/( kg·d-1 ) ) ,and there were 12 mice in each group. All the mice in the different groups were given the corresponding agents orally and those in the control group were given the same volume of distilled water once a day continuously for 15 days. After the last administration,locomotor activities and standing times of mice in all the groups were measured by a multiple functional mice locomotor activities recorder, and the threshold dose pentobarbital sodium-induced sleep experiment was conducted, to examine the sedative and hypnotic effect of WShC. The contents of 5-Hydroxytryptamine (5-HT) ,5-Hydroxyindole acetic acid (5-HIAA) ,γ-aminobutyric acid ( GABA) and glutamine ( Glu) in the hypothalamus were detected with ELISA method for exploring the underlying mechanisms. Results Compared with the control group,the locomotor activity was reduced,the sleep latency was shortened and the sleep duration was prolonged in all the WShC-treated groups ( P<0. 01 ). WShC could significantly increase the contents of 5-HIAA and 5-HIAA/5-HT proportion;decrease the contents of Glu and Glu/GABA proportion in the hypothalamus. Conclusion WShC has a sedative and hypnotic effect on mice, which might be related to the regulation of central excitatory and inhibitory neurotransmitters.关键词
