Role of ventricular M2 receptors expression on the effects of remifentanil in ventricular transmural repolarization heterogeneity

Abstract
Objective To investigate the role of ventricular M2 receptors expression in the effects of remifentanil on ventricular transmural repolarization heterogeneity.Methods Twenty-four adult rabbits were randomly divided into four groups after successful preparation of Langendroff isolated heart perfusion model (n =6):control group was perfused with K-H solution for 60 min,low,middle and high test groups were respectively perfused with 12,25,50 ng · mL-1 remifentanil K-H solution for 60 min.Transmural dispersion of repolarization (TDR) was recorded at the time of reperfusion 60 min.The expression of M2 receptors protein in the ventricular myocardium was determined by Western blotting.Results The TDR of the control group was 8.42 ± 5.33,and the TDR of low,medium and high concentration test groups were 41.24 ± 15.59,29.64 ± 12.40,31.13 ±20.88,with significant difference (P < 0.05).The expression of M2 receptor in control group,low concentration and middle concentration test groups were 0.32 ±0.02,0.40 ±-0.01,0.45 ±0.03,and the expression of M2 receptor in high concentration test group was 0.63 ± 0.04,with significant difference (P < 0.05).Conclusion Remifentanil dose-dependently up-regulates the expression of ventricular M2 receptor,which may be the mechanism of ventricular transmural repolarization heterogeneity.关键词
