金雀异黄素对雌性大鼠体内促性腺激素及胰岛素样生长因子表达的影响
Effect of Genistein on Expressions of Gonadotropins and Insulin?Like Growth Factors of Female Rats

Abstract
In order to study the effects of genistein ( GEN ) on expressions of gonadotropins and insulin?like growth factors in female rats, forty female SD rats [(200±20) g] were selected, and randomly divided into 5 groups with 8 rats per group, which were negative control ( NC) group, GEN low dose ( L) group, GEN middle dose ( M) group, GEN high dose ( H) group, positive control ( PC) group. Rats in NC group were in?tragastric administrated peanut oil ( other groups used it as solvent) , those in L, M and H groups were intra?gastric administrated GEN at 15, 30 and 60 mg/( kg BW·d) , and those in PC group were intragastric admin?istrated diethylstilbestrol at 0.5 mg/( kg BW·d) . The experiment lasted for 30 days. Serum contents of folli?cle?stimulating hormone ( FSH) , luteinizing hormone ( LH) , insulin?like growth factor?1 ( IGF?1) and insu?lin?like growth factor binding protein?1 ( IGFBP?1 ) were detected by enzyme linked immunosorbent assay ( ELISA) method. The mRNA expression levels of IGF?1 and IGFBP?1 in ovary tissue were detected by real?time quantitative PCR. The results showed as follows: compared with NC group, serum contents of FSH and LH in experimental groups tended to be increased, but the differences were not significant ( P>0.05) , which showed the same tendency with those in PC group; serum content of IGF?1 in experimental group was slightly decreased, but the difference was not significant ( P>0.05) , and that in PC group was significantly decreased (P<0.05); serum content of IGFBP?1 in experimental groups was significantly increased (P<0.05 or P<0.01), and that in PC group was significantly increased (P<0.05); mRNA expression levels of IGF?1 and IGFBP?1 in ovary tissue were increased, especially were significantly increased in M and H groups ( P<0.05) , which showed the same tendency with those in PC group. It is concluded that GEN can increase serum contents of FSH and LH, decrease serum IGF?1 content, and increase serum IGFBP?1 content, as well as increase mR?NA expression levels of IGFBP?1 and IGF?1 in ovary of rats. These indexes synergistically affect ovary, can promote follicle maturation and improve the function of ovary.关键词
