The effects of KCl stimulation on the expression of MKK4 in glioblastoma cells

Abstract
Objective To observe the expression of MKK4 and its downstream protein p38 MAPK after stimulating human astrocytoma cells (U87) with different concentrations of KCl,and to explore the expression of MKK4 and p38 MAPK at the level of nucleic acid.Methods U87 cells were stimulated with different concentrations of KCl,the expression of MKK4 and p38 MAPK mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR).The cells were divided into 5 groups:control group (group 0),different concentrations of KC1 group (0.001 mmol/mL,0.025 mmol/mL,0.05 mmol/mL,0.1 mmol/mL,labeled 0.001,0.025,0.05,0.1 group).Results The results of RT-PCR showed that MKK4 and p38 MAPK mRNA were expressed in astrocytes after stimulation with KCl,the expression was increase and then decrease and in the 0.05 group reached the peak,then fall.Conclusion KCl stimulation can increase the expression of MKK4 and p38 MAPK mRNA in human astrocytoma cells,and its expression is related to the concentration of KCl.关键词
