影响鸡传染性法氏囊病毒抗原(HQ株)效价因素的探讨
Exploration on Antigen Titer Factors Affecting Chicken Infectious Bursal Virus (HQ Strain)

Abstract
Antigen titer is an important indicator in the evaluation of vaccine quality.Currently the bursal virus vaccines are not so effective,and one of the reasons is the less effective antigen titer.In this experiment,the influencing factors for infectious bursal virus antigen (strain HQ) titer in chicken were explored so as to optimize production technological parameters and improve antigen titer,therefore improving the immunity effect of bursal virus vaccines.The best production process parameters were determined through comparing the cell density,pH value of cell maintaining fluid,concentration of virus,transmitting time,infection time,culture temperature after infection and the serum concentration of maintaining fluid.Results show that dense cell could improve the antigen titer.The pH value of cell maintaining fluid was 7.8 that could improve the antigen titer compared with pH value 7.4.The antigen titer was the highest in 1 % virus concentration.The antigen titer was not significantly different with the cell cultured for 24h and 48h.The best time of harvested virus was 36 hours after infection.And the best culture temperature of cell after infection was 37℃.The antigen titer was the highest in 2% serum in cell culture fluid.So the best production process parameters of the chicken infectious bursal disease vaccine production were obtained.关键词
