Effects of sericine on interferon-γ,tumor necrosis factor-α, interleukin-4 ,interleukin-10 in the spleen of type Ⅱ diabetic rats

Abstract
Objective:To investigate the effects and possible mechanisms of sericine on immunologic function of type Ⅱ diabetic rats by detecting changes of interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),interleukin-4 (IL-4)and interleukin-10 (IL-10)expression in the spleen.Methods:The type Ⅱ diabetic rat model was established by continuous peritoneal injection of small-dose streptozotocin.Rats were randomly divided into four groups,normal control group,type Ⅱ diabetes model group,sericine treatment group,and melbine treatment group.Rats in sericine treatment group and melbine treatment group were lavaged with sericine and melbine for 35 days.Glucose oxidase method was used to detect the blood glucose of rats,and Western blotting and RT-PCR were used to detect the expression of IFN-γ,TNF-α,IL-4 and IL-10 protein and mRNA in the spleen.Results:Compared with rats in the normal control group,blood glucose,IFN-γand TNF-αexpression were obviously increased in the model group.IL-4 and IL-10 expression decreased remarkably in the spleen of rats in the model group. Compared with the rats in the model group,IFN-γand TNF-αexpression were decreased obviously,and IL-4 and IL-10 expression were increased obviously in the spleen of rats in the sericine treatment group.Conclusion:Sericine can regulate the immune function of type Ⅱ diabetic rats by down-regulating IFN-γ and TNF-αexpression,and up-regulating IL-4 and IL-10 expression in the spleen.关键词
