摘要

Abstract

Objective:By co-culturing the mixture of the bone marrow mesenchymal stem cells (BMSCs) with primary articular chondrocytes (AC) coaggulated in the centrifuge tube,efficacy of chondrocytes induction was compared between co-culture and chondrocytes-only culture.Methods:After BMSCs and AC were isolated from the marrow and cartilage of New Zealand white rabbits,BMSCs and AC were cultured in vitro separately.Alcian blue,safranin-O staining and type Ⅱ collagen immunohistochemical staining were applied to identify chondrocytes.Expressions of CD44,CD105,CD34 and CD45 were immunofluorescencally assessed to identify BMSC.After culturing,the primary chondrocytes were mixed with the third generation of BMSCs by 1:3 in the centrifugal tube,as co-culture group.Meanwhile,the pure AC cultured as the control,as AC-alone group.Cell supernatants were collected weekly for 4 weeks and glycosaminoglycan(GAG) and type Ⅱ collagen in two groups were determined.Results:There was no significant difference of gross morphology and histological observation in two groups.The ability of secreting GAG and type Ⅱ collagen were also similar in two groups(P > 0.05).Conclusion:co-culturing BMSCs with chondrocytes in the centrifuge tube could promote induction of BMSC differentiating into AC successfully.
