Abstract
Objective:To study of the association between polymorphism of the MMP-9 gene(-1562C/T)and type 2 diabetic retinopathy. Methods:MMP-9-1562C/T SNP was genotyped by polymerase chain re-action-restrictive fragment length polymorphism ( PCR-RFLP ) ,analysis in 180 healthy persons as control and 400 patients with type 2 diabetes including 180 complication-free cases and 220 with diabetic retinopathy ( DR) . Results:Association between the MMP-9 -1562C/T SNP in promoter region and diabetic retinopa-thy:the C and T allele frequencies in Control,DM,DR groups are 8.3%,91.7%;7.8%,92.2%;91.4%,8. 6% respectively. And the results have a significant deviation (χ2=790.09,P=0.000) . The C/C,C/T,T/T genotypic frequencies of cases in the three groups are 0%,16.7%,83.3%;2.2%,11.1%,86.7%;84.1%,14. 5%,1.4%.There's a significant deviation of C and T allele frequency of MMP-9 -1562C/T between the Con-trol group,the DM group and DR group(χ2=470.16,P=0.000).There's a significant deviation of C/C,C/T,T/T genotypic frequencies between the Control group and DM group( P<0.01),or the DM group and DR group (P<0.01),while there is no significant deviation between the Control group and DM group(P>0.01) .Com-pared with the T allele, the C allele may increase the onset risk of diabetic retinopathy. Conclusion: The MMP-9 -1562C/T SNP may play a role in the genetic susceptibility to the invasion of diabetic retinopathy. T allele is a protective gene for diabetic retinopathy.关键词
