摘要

Abstract

Objective:To investigate the capacity transforming growth factor-β3 ( TGF-β3 ) to induce rabbit dental pulp stem cell ( DPSCs) osteogenic differentiation in vitro. Methods:DPSCs were isolated with tissue enzymetic digestion method to observe the morphology under light microscope. The four-passage DPSCs cultured in vitro were divided into three groups,and incubated respectively with total medium( control group) ,10 mmol/L β-glycerophosphate,10 mol/L dexametha-sone and 50 mg/L vitamins C(mineralized solution positive control group),and 80 ng/mL TGF-β3(experience group). Al-kailine phosphate(ALP)activity assay was adoted to detect ALP activity after 1,3,5,7 days. Immunocytochemistry assay was adopted to detect the expressions of related transcriptionfactor 2(RUNX-2)and bone sialoprotein(BSP)after 1,3,5,7 days. Alizarin red staining was adoted to observe the mineralized nodules after 14 days. The effect of TGF-β3 on the osteogenic was observed by Western blot. Results:DPSCs grew well in inducing system. Compared with control group,the ALP activity was up-regulated. Runx-2 showed positive by 5 days,and BSP was positive by 7 days after ostegenic induction in TGF-β3 and mineralized solution groups,and both RUNX-2 and BSP were weakly positive in blank control group. Alizarinred staining was positive by 14 days in mineralized solution group and was strong positive in TGF-β3 group. Western blot revealed that the os-teogenic marker RUNX2 was significantly upregulated in cells compared with the control group. Conclusion:TGF-β3 acceler-ates the osteogenic differentiation of DPSCs.
