Expressions and Clinical Significances of OX40, OX40L, PD-1, PD-L1 and SPD-1 in Peripheral Blood of Patients with Primary Sjogren's Syndrome

Abstract
Objective To investigate expressions and clinical significances of OX40, OX40L, PD-1, PD-L1 and SPD-1 in peripheral blood of patients with primary Sjogren's syndrome ( PSS) . Methods A total of 50 patients with PSS ( ob-servation group) admitted during March 2012 and March 2015 were divided into active period subgroup (n=30) and inactivi-ty period subgroup (n=20) according to disease activity conditions, and single system subgroup (n=29) and multisystemic subgroup (n=21) according to active stages. Other 50 healthy people taking medical examinations at the same period were selected as control group. OX40, OX40L, PD-1, PD-L1 and SPD-1 expressions in peripheral venous blood were detected by immunofluorescence labeling and flow cytometry methods in two groups. OX40, OX40L, PD-1, PD-L1 and SPD-1 expressions were compared in groups and subgroups, and changes of the above indexes before and after treatment were recorded in observa-tion group. Results In observation group, CD4 +OX40 +, CD14 +OX40L+, CD19 +OX40L+, CD4 +PD-1 + and CD4 +PD-1L+ and SPD-1 expressions were significantly higher, while CD14 +PD-L1 + was significantly lower than those in control group (P<0. 01). CD4 +OX40 +, CD14 +OX40L+, CD19 +OX40L+, CD4 +PD-1 + and SPD-1 expressions in active period sub-group were significantly higher than those in inactivity period subgroup ( P <0. 05 or P <0. 01 ); CD4 +OX40 +, CD14 +OX40L+, CD19 +OX40L+ and SPD-1 expressions in single system subgroup were significantly lower than those in multisys-temic subgroup ( P<0. 05 or P<0. 01 ) . In 10 preliminary diagnosis patients in observation group, CD4 +OX40 +, CD14 +OX40L+, CD19 +OX40L+ and CD4 +PD-1 + expressions before treatment were significantly higher than those after treatment ( P<0. 05);there were no significant differences in CD8 +PD-1 +, CD14 +PD-L1 + and SPD-1 expression between before and after treatment (P>0. 05). Conclusion Abnormal expressions of OX40, OX40L, PD-1, PD-L1 and SPD-1 in peripheral venous blood of PSS patients relate to disease activity and tissue damage, and the expressions can be used as detection indexes for learning disease activity conditions and severe degree.关键词
