Abstract

Objective:To investigate the effects of miR-425 regulates the proliferation and apoptosis of renal epithelial cell by regulating PDL-1 pathway.Methods:Nasal polyp epithelial cells HRCEpiC were transfected miR-425 mimic and control,qRT-PCR method was used to detect the expression level of miR-425,MTT method and Tunel test were used to detect the cell proliferation and apoptosis,the level of PDL-1 expression were determined by Western blot experiment.Results:The results of qRT-PCR showed that the expression level of miR-425 in the ex-perimental group was about 37.82 times higher than that in the blank group and the negative control group.After transfection of 48h,the proportion of live cells in the experimental group,negative control group and blank group were(75.22 ± 3.19)%,(98.29 ± 2.11)%and(100 ± 0.00)%,the apoptosis rate were(17.20 ± 2.49)%,(4.29 ± 1.44)%and(0.52 ± 0.11)%,there were statistically significant difference compared among the groups(P<0.05);Western blot assay showed that transfection after 48 h,the expression level of PDL-1 in the experimental group were significantly lower than the negative control group and blank control group(P<0.05).Conclusion:miR-425 can in-hibit the proliferation of nasal polyps epithelial cells by increasing PDL-1 pathway,and improve the ability of apopto-sis.
