摘要

Abstract

Objective:To detect the expression level of 5-LOX in ovarian cancer,ex-plore the correlations between 5-LOX expression and clinicopathological features, and investi-gate its effect on the development and treatment of ovarian cancer. Methods:The expressions of 5-LOX and HIF-1α in 56 ovarian cancer specimens was examined by immunohistochemistry, and the correlations between 5-LOX expression and clinicopathological features were further an-alyzed. The human ovarian cancer SKOV3 cells were cultivated 0h,12h,24h,48h respectively under hypoxia environment, and then RNA and protein were extracted. Quantitative real-time PCR ( qRT-PCR) was used to measure the mRNA expression level of 5-LOX. Western blot was used to measure the protein expression level of 5-LOX. ELISA was used to measure the change of 5-HETE. Result:Immunohistochemistry results showed that 5-LOX level was correlated with HIF-1αlevel,and its level was significantly correlated with lymph nodes metastasis,distant me-tastasis and clinical stage. 5-LOX was up regulation both in transcription and protein levels in o-varian cancer SKOV3 cells under hypoxia. The 5-HETE level was also increased under hypoxia. Conclusions:Hypoxia can promote the expression of 5-LOX and increase its metabolites 5 -HETE in ovarian cancer,suggesting 5-LOX can be a potential target for the treatment of ovarian cancer,which provides a new thought and method in the pathogenesis and treatment for ovarian cancer.
