Study on the mechanism of PTEN-impaired trophoblast migration in preeclampsia

Abstract
Objective:To investigate the expression levels of PTEN in placental sam-ples from patients with severe preeclampsia ( PE) and normotensive pregnancies. To study the underlying mechanism of regulation of trophoblast migration by PTEN. Methods:Twenty cases of placental samples were collected from patients with severe PE and normotensive pregnancies respectively. Quantitative real-time PCR and Western blot were used to detect the expression of PTEN in the placenta. HTR-8/SVneo cells were transfected with pcDNA3. 1-HA-PTEN and siPTEN respectively,pcDNA3. 1 and siCTL as controls. Wound healing assays were performed to assess cell migration as well as Western blot was used to detect the expression levels of p-Akt (Thr308),p-Akt (Ser473),Akt,MMP-2 and MMP-9. Results:The mRNA and protein levels of PTEN were both significantly higher in severe PE placenta compared with the control group (P<0. 01). After the HTR-8/SVneo cells were transfected with pcDNA3. 1-HA-PTEN for 48h, the migration rates were significantly reduced [ ( 26 . 42 ± 6 . 68 )% vs ( 52 . 92 ± 6 . 71 )%, P<0. 01],and the expression of p-Akt (Thr308) and p-Akt (Ser473) significantly decreased,as compared to that of the cells transfected with pcDNA3 . 1 , while Akt had similar expression in these groups. Furthermore,overexpression of PTEN in HTR-8/SVneo cells reduced the levels of MMP-2 and MMP-9 protein. In contrast,the migration rates of HTR-8/SVneo cells transfected with siPTEN for 48 h were significantly increased[(50. 39±6. 84)% vs (30. 04 ±5. 40)%,P<0. 01],and p-Akt (Thr308) and p-Akt (Ser473) expression significantly increased compared with that of the cells transfected with siCTL,whereas the expression of Akt was not significantly different in these groups. And,knockdown of PTEN in HTR-8/SVneo cells increased the levels of MMP-2 and MMP-9 protein. Conclusion:The expression of PTEN was elevated in severe PE placenta,and PTEN inhibits trophoblast migration via down-regulating the activity of Akt and its downstream targets MMP-2 and MMP-9 expression,indicating that PTEN may play an important role in the pathogenesis of PE.关键词
