The effect of stanniocalcin-1 on the malignant transformation of ovarian epithelial cells

Abstract
Objective:To study the effect of STC1 on the malignant transformation of o-varian epithelial cells. Methods:STC1 gene was transfected into immortalized ovarian epithelial cells(T29-STC1)by lentivirus transfection as study group,while control group(T29-Vector) was transfected with empty vector. Western blot was used to detect the expression of STC1 in T29-STC1 cells and T29-Vector cells. Cytometry was used to determine whether the level of STC1 expression affects cell cycle progression. Plate cloning experiments was used to count the number of cell formation after disposed by carboplatin. MTT assay was used to detect the prolif-eration ability of the cells and subcutaneous tumor xenograft in nude mice was used to detect the ability of tumor formation. Results:Western blot showed that the expression of STC1 in T29-STC1 was significantly enhanced. Cytometry showed that the T29-STC1 cells in G0 and G1 cell cycle decreased but increased in S cell cycle in comparision with T29-Vectorcells. Plate cloning experiments and MTT showed that the growth rate of T29-STC1 was significantly higher than that of T29-Vector after carboplatin treatment. T29-STC1 inoculated in nude mice could form tumor while T29-Vector could not. Conclusion:Stanniocalcin-1(STC1)protein may be involved in malignant transformation of ovarian epithelial cells.关键词
