Abstract

The aim of this study was to study the effect of mulberry leaf tea on MDA-MB-231 cell proliferation,apoptosis,cell cycle and Survivin expression,and to explore its mechanism.MDA-MB-231 cells were cultured in vitro.The effect of mulberry leaf tea on cell proliferation was detected by CCK8 method.The apoptosis of MDA-MB-231 cells was analyzed by Hoechst33258 staining and Rhodamine 123 staining.The effect of cell cycle was analyzed by flow cytometry.The effect of mulberry leaf tea on Survivin protein expression was detected by Western Blot blot.The results showed that the growth of MDA-MB-231 cells was significantly inhibited by the mulberry leaf tea,the cell proliferation was inhibited and the number of apoptotic cells increased significantly.The G0/G1 phase cells of MDA-MB-231 cells were significantly higher than those in the normal group.The expression of Survivin protein in the mulberry leaf tea group was lower than that in the control group,and the expression level decreased with the increase of mulberry leaf concentration.Hence,it was concluded that mulberry leaves tea can inhibit the proliferation of MDA-MB-231 cells,promote cell apoptosis and lead to G0/G1 arrest and decrease the expression of Survivin.
