松脂醇二葡萄糖苷对UVB诱导HDF细胞损伤的保护作用及机制研究
Study on Protctive Effects and Mechanism of PDG on UVB Induced HDF Cells Damage

Abstract
Objective:To investigate the protective effect and the mechanism of PDG on UVB induced HDF cells injury.Methods:The rate of cell proliferation was determined by MTT assay,the collagen protein in cells,matrix metalloproteinases-1 (MMP-1),ERβ,p-P38 protein expression level were measured by Western blot,and the expression of MMP-1 mRNA was detected by real-time fluorescent quantitative PCR method.Results:Compared with the control group,the cell proliferation rate and ERβ,COL Ⅰ protein expression in model group decreased obviously,MMP-1 and p-P38 protein expression and MMP-1 mRNA expression increased significantly (P <0.01).Compared with the model group,cell proliferation rate and collagen of PDG group increased significantly.At the same time,PDG inhibited the expression of MMP-1 protein and MMP-1 mRNA,and up-regulated the expression of ERβ protein and decreased p-P38 protein expression significantly(P < 0.01).Compared with the PDG group,ICI182780 was significantly suppressed ERβ protein expression and up-regulated MMP-1 mRNA expression,and SB203580 significantly inhibited the expression of MMP-1 and p-P38 protein,and decreased mRNA expression of MMP-1 (P < 0.01).Conclusion:The results indicated that PDG can significantly promote the proliferation rate of HDF cells,and mechanism may through the ER-P38MAPK signaling pathway inhibiting expression of MMP-1 protein and MMP-1 mRNA,to promote the synthesis of collagen to achieve the purpose of prevention and treatment of light aging.关键词
