Application of DH-PSF three-dimensional dynamic multi-particle tracking method in living cell imaging
摘要
Abstract
Multi-particle tracking is one of the most challenging aspects of living cell imaging.We establish a three-dimensional multi-particle tracking imaging system based on three-dimensional positioning of double helix point spread function.The imaging depth and localization precision can achieve 4 μm and 20 nm,respectively.The result of dynamic tracking of fluorescence microspheres in Raw cells shows that the detectable trajectory range of intracellular moleculesis could be up to 7 μm and the velocity of probes could be obtained in real time.Therefore,this system is a very useful tool for studying the intermolecular interaction in living cell.关键词
