杏鲍菇菌糠漆酶分离纯化及部分酶学性质研究
Purification and Properties of Laccase from Spent Mushroom Compost of Pleurotuseryngii

Abstract
The main aim of this study was to finda cheap raw material,spent mushroom compost of Pleuro-tuseryngii,which can be used to extract laccase.Laccase from mushroom dregsof P.eryngii was graded-precipi-tated with ammonium sulfate,frozen and dried,separated and purified with chromatography on DEAE-cellulose and its enzymatic properties were studied.The resuts showed:the laccase detected by Native SDS-PAGE was a single pure product,its molecular weight was about 44.3 ku,the purification of about 10.452-fold was achieved with an overall yield of 16.640%;the specific activity of the laccase was 6.233 U/mg,the Mc value of its oxi-dation of ABTS was 0.0870 mmol/L;the optimum temperature for laccase activity was 40℃,and the laccase could work continuously under 35℃;the optimum pH for the laccase activity was 3.8,and this laccase showed a good stability when the pH of the buffer varied from 3.8 to 4.6;the laccase activity was enhanced by Fe2+, Cu2+ and was strongly inhibited by Fe3+,Mn2+.关键词
