PRP对大鼠腹部岛状皮瓣缺血再灌注损伤的影响
EFFECT OF PLATELET-RICH PLASMA ON ISCHEMIA-REPERFUSION INJURY OF THE ABDOMINAL ISLAND FLAPS IN RATS

Abstract
Objective To investigate the effect of platelet-rich plasma (PRP) in the treatment of ischemia-reperfusion injury of the abdominal island flaps in rats.Methods A rat model of ischemia-reperfusion injury of the abdominal island flaps was established.The rats in experimental group were given PRP injection via the contralateral femoral artery,those in control group were given the injection of normal saline via the contralateral femoral artery,and those in blank group were not given vascular occlusion or medication after flap preparation.Blood flow volume in the flap and the content of malondialdehyde (MDA) and nitric oxide (NO) in the flap tissue were measured at different time points of ischemia and reperfusion.At 7 d after in situ flap suture,the designed area and survival area of the flap were measured and the survival rate of the flaps was calculated.Immunohistochemical staining was performed for flap tissue to measure the expression of vascular endothelial growth factor (VEGF).Results At 2 h after ischemia and 30 min and 2 h after reperfusion,there was a significant difference in the blood flow volume of the microcirculation in the abdominal island flaps between the three groups (F=16.85-35.02,q=3.64-11.54,P<0.05).At 5.5 and 7.0 h after surgery,there were significant differences in the content of NO and MDA in the flap tissue between the three groups (F=3.87-605.40,q=3.76-48.02,P<0.05).At 7 d after surgery,there was a significant difference in the survival rate of the flaps between the three groups (F=339.60,q=11.38-36.05,P<0.05).Conclusion PRP exerts its protective effect against ischemia-reperfusion injury by increasing the content of NO,reducing the level of MDA,increasing the expression of VEGF,and improving neovascularization and flap microcirculation.关键词
