Abstract

The enzyme linked immunosorbent assay (ELISA) is a rapid diagnostic method,and is convenient for clinic practice.In this study,the N protein prokaryotic expression of PEDV AH2012 strain was established previously.And recombination N protein has been expressed.Furthermore,indirect ELISA method was established for the detection of IgG antibody,and each steps of method were optimized.The result showed this indirect ELISA was a stable,accurate method,and rarely influenced by environment factors,such as temperature.Test results indicated this ELISA method was sensitive and specific,and there were no cross-reactions with PRRSV,PRV,TGEV,FMDV,PCV2,PRCV positive sera.The intra-and inter-assay coefficients of variation were lower than 5.55%.The coincidence rate of results of the ELISA and the neutralization test was 88.75%.The ELISA method could be applied to the diagnosis of PEDV,PEDV epidemiological surveillance and vaccine antibodies detection.This ELISA method is a promising candidate for the detection and prevention of PEDV.
