肠炎沙门菌感染ICR小鼠模型的建立
Establishment of a ICR mouse model infected with Salmonella enteritidis

Abstract
Salmonella enteritidis(SE)is one of the most important Gram-negative bacteria that are human food-borne pathogens,which can cause serious symptoms of infection in animals such as poultry and livestock.In order to create a ICR mice model infected by SE,the standard strain SE50336 were inoculated orally into 6-weeks-old ICR mice groups (each group containing 10 mice) with the dose of perfusion 1 × 108,1 × 107,1 × 106,1 × 105 and 1 × 104 CFU per mouse,respectively.The other 10 mice were inoculated by PBS as a control group to measure LD50 of SE-infected ICR mice.Another 40 mice were divided into two groups by gender,and inoculated with the dose of calculated LD50 of SE.Clinical symptoms,changes of body weight,changes of histopathological,tissue bacterial load and the mRNA relative expression levels of TNF-α IL-6 were evaluated for the pathogenicity of SEto mice.In result,the LD50 of SE50336 was 1 × 105.67 CFU.After 3 days the infected mice showed feed intake decreasing,activity reducing,skin color darker,chills,gathered and so on;the average weight of the mice infected on day 5 post-infection was significantly decreased,and the latter began to recover.The histopathology analysis suggested that the liver had appeared thrombosis and necrosis with bacterial clumps in parenchyma;the spleen mainly showed the cellulosic necrotizing spleen inflammation,with white intramedullary lymphopenia decreased.Colony loading showed that the bacterial load increased with the infection process,and the bacteria in the spleen was the highest,followed by the spleen.Quantitative PCR results showed that the mRNA expression of TNF-α was significantly increased from day 2 to 8 postinfection.However,IL was significantly decreased from day 4 to 8 post-infection.The above-mentioned experimental results showed that the ICR mice could be utilized as a SE infection model.This study provided a valuable animal model for the study of SE infection and pathogenesis,and also laid the foundation for evaluating the anti-SE evaluation of antimicrobial peptide transgenic mice.关键词
