Effects of transforming growth factor-β1 on the expression of fibroblast activation protein in oral carcinoma-associated fibroblasts

Abstract
Objective To explore the effect of transforming growth factor-β1 (TGF-β1) on the expression of fibroblast activation protein (FAP) in carcinoma-associate fibroblasts (CAFs).Methods CAFs cultured in vitro were treated with the TGF-β1 (10 ng/ml).The normal fibroblasts(NFs) and untreated CAFs cells as control groups,Western blot analysis and reverse transcription polymerase chain reaction(RT-PCR) are employed to detect the expression of FAP mRNA and protein.Results The results displayed that FAP was undetectable in NFs group,but the expression of FAP mRNA and protein increased in CAFs group.Compared with the control groups,FAP was significantly upregulated after treatment with 10 ng/ml TGF-β1 in oral CAFs cells (P ＜ 0.05).The expression of FAP increased continuously at 24 h compared with that at 12 h,and the difference was statistically significant.Conclusion TGF-β1 can facilitate the FAP expression in CAFs,and may play important roles in the interaction between epithelial cells and adjacent mesenchyme.关键词
