栎树桑黄提取物诱导胃癌细胞凋亡作用的研究
Study on the apoptosis of gastric cancer cells induced by extractions from oak Phellinus linteus

Abstract
Objective To investigate the antitumor and apoptosis induction effect of extractons from oak Phellinus linteus(OPL) against human gastric SGC-7901 cancer.Methods Human SGC-7901,HCT-116,MCF-7,Hela cancer cells were cultured with OPL(0,40,80,160,240,320,400 μg/ml) for 48 h.The proliferation was evaluated via MTT assays,the cell cycle was analyzed through flow cytometry,Hoechst staining was used to observe OPL impact on nuclear shape change,and the apoptosis state was analyzed by Annexin-V/PI staining assays.The expression of apoptosis-related and cycle-related proteins was determined by Western blot.Results OPL could significantly reduce cell viability,induce apoptosis,and arrest SGC-7901 cell cycle at the G0/G1 phases(P ＜0.05).Western blot method further confirmed that OPL could increased bax and the subsequent cleavage of poly (ADP-ribose) polymerase,decreased CyclinD1 and Bcl-2 protein expression (P ＜ 0.05).Conclusion Our findings suggest that OPL can induce apoptosis in human gastric SGC-7901 cancer cells,and cause cell cycle arrest in G0/G1 phase.关键词
