独正杆接骨膏对家兔骨折愈合过程中CHORDIN和NOGGIN mRNA表达的影响
Effect of Duzhenggan Jiegu ointment on expression of CHORDIN and NOGGIN mRNA in process of fracture healing
摘要
Abstract
Objective To investigate the Duzhenggan Jiegu ointment on rabbit fracture healing process CHORDIN and NOGGIN mRNA expression, and analyze themechanism of promoting fracture healing. Methods 30 rabbits were randomly divided into experimental group and control group, 15 rabbits in each group. In experimental group rabbits skin external application of Jiegu ointment and fixed with small splints;The control group uses no medicine but routine disinfection and small splint fixed. 5 rabbits were randomly selected from each group at third, sixth, tenth weeks after the administration. The rabbits were sacrificed and the tissue samples were taken. The expression of CHORDIN and NOGGIN mRNA in bone tissue of rabbits was measured by real-time fluorescence quantitative PCR method, and the data were statistically analyzed. Results Compared with the control group, the expression of CHORDIN, NOGGIN and mRNA in the bone tissue of rabbits in the experimental group was significantly decreased at third weeks, the difference was statistically significant (P<0.05); The expression of CHORDIN, NOGGIN and mRNA in bone tissue of rabbits in sixth weeks was not statistically significant compared with the control group (P>0.05); The expression of CHORDIN, NOGGIN and mRNA in the bone tissue of rabbits in tenth weeks was significantly higher than that in the control group,the difference was statistically significant(P<0.05).Conclusion Duzhenggan Jiegu ointment may inhibit CHORDIN, NOGGIN mRNA expression in early and late expression, so as to promote the normal conduction of normal BMP signaling pathway, the osteogenic differentiation of BMSCs, to promote the healing of bone fracture.关键词
