摘要

Abstract

Proline accumulation has been proven to play protective roles in response to different environmental stresses.A1-pyrroline-5-carboxylate synthetase 1 (P5CS1) is the key enzyme for proline biosynthesis,especially for proline accumulation under the condition of stresses.To investigate the possible mechanisms of P5CS1 and proline,we developed a P5CS1 over-expression construct and transformed it into Arabidopsis.We also developed a P5CS1 prokaryotic expression construct for further functional study.The results showed that,over-expression of P5CS1 gene significantly enhanced proline accumulation under both normal growing and salt stress treatment conditions,and then improved plant tolerance to salt in either medium or soil.The prokaryotic expression revealed that,P5CS1 protein had higher expression in condition of 28 ℃C with 0.8 mmol · L-1 IPTG,and mainly expressed in the inclusion body of E.coli.These results indicate that P5CS1 is the key enzyme for proline accumulationin Arabidopsis,and has important roles in protecting plants in condition of salt stress.The fusion protein 6 × His-P5CS1 expressed here lays a basis for further investigating the molecular mechanism of proline in regulating plant tolerance to stresses.
