摘要

Abstract

Objective To investigate the effects of hepatitis B virus X protein A1762T/ G1764A double mutation on the expression of clock gene in the hepatocellular carcinoma (HCC)cells. Methods Bel-7404 cells were inoculated into into 6-hole plate,when the cells grew to 60% confluence,they were transfected with 1. 0 μg pcDNA3. 1-HBx and pcD-NA3. 1-mHBx (A1762T/ G1764A double mutant)plasmids by using Lipofectamine 2000 according to the manufacturer's in-structions. The mRNA and protein expression levels of the CLOCK,BMAL1,Per1,Per2,Per3,Cry1,Cry2,and CKIεgenes in the Bel-7404 cells were detected by real-time PCR and Western blotting. Results The mRNA and protein expres-sion of Clock,Bmal1,Per2,Per3,Cry1 and Cry2 decreased in Bel-7404 cells transfected with A1762T/ G1764A double mutant HBx vectors as compared with that of cells transfected with pcDNA3. 1-HBx vectors,and the differences were statis-tically significant (all P < 0. 05). Conclusions HBx A1762T/ G1764A double mutation can cause the disorder of biologi-cal clock expression in HCC cells.
